Abstract : Objective: This work was conducted to prepare and characterize a Modafinil-nicotinic acid cocrystal loaded mouth dissolving film (MDF) through formulation-optimization approach.  Methods: Modafinil and nicotinic acid were mixed in a 1:1 molar ratio and characterized in terms of aqueous solubility, FTIR, powder x-ray diffraction (PXRD) and SEM analysis. The cocrystal was prepared in HPMC E5 films by solvent casting method. Variables including HPMC E5, PEG 400 and crospovidone were studied using three-factor three level BoxBehnken design with disintegration time as the main response variable. Optimized MDF was evaluated in terms of appearance, weight variation, folding endurance, surface pH, drug content, disintegration time, in-vitro dissolution and release kinetics.
